Underwater Visual Census videos at pilot oyster reefs in the Borkum Reef Ground marine protected area (HEINCKE cruise HE601)

The Underwater Visual Census (UVC) transects were carried out by scientific divers during the Heincke HE601 cruise focusing on developing and testing monitoring techniques at the Borkum Reef Ground and Sylt Outer Reef Marine Protected Areas. The UVC transects consisted of 1x1m sampling regions. To define these regions, divers used a 1m-long PVC pipe. At each sampling region, the diver spent approximately 1 minute counting and identifying, to the lowest taxonomic level possible, as many organisms as possible. The UVC transect provide insights into the general composition of key species, higher systematic groups and ecological guilds. Seabed images taken at pilot oyster reefs in the Borkum Reef Ground marine protected area during September 2021

Imagery transects with a camera attached to a frame design for taking pictures in a 50 x 50 cm area. Seabed images transects were carried out by scientific divers during the RESTORE expedition on board the ship Arctic Ocean during September 2021, focusing on the monitoring of two pilot reefs (east and west) and the ecology of benthic assemblages in the Borkum Reef Ground marine protected area. The camera used was a Cannon EOS M6 camera, the pictures were taken perpendicularly to the seabed. The transects were done along a preset track across the reefs, number of pictures per transect varied based on diving time, with a minimum of 10 images per transect. Due to logistic problems, two transects were done for the east reef, and one for the west reef. The seabed images provide insights into the general composition of key species, higher systematic groups and ecological guilds. The images also contain valuable information on how benthic species are associated to each other. Dataset includes individual images, and metadata of each image including diving depth, date/time, and corresponding diver. Detailed geographical coordinates of individual images are unavailable.

Results of calcein staining with three different concentrations and immersion times on the growth of Ostrea edulis

Chemical dyes, such as Calcein, can be used to create incremental markings for measuring bivalve growth in shell cross-sections. For the European Oyster Ostrea edulis, potential negative effects and limits for this staining have not been assessed yet. In this study, Ostrea edulis individuals were stained with three different Calcein concentrations (100 mg l−1, 150 mg l−1, 200 mg l−1) at three different immersion times (6 h, 12 h, 24 h) to test the potential negative effects, the best incremental marking result and their potential to be used in growth measurements. The dataset includes the growth measurements of the individuals at the time of staining in Mai 2021 and the growth measurements after a growth period of 5 months (N=10 per treatment). In November 2021, the success of the staining was evaluated using a fluorescence microscope by measuring the increment from the staining line to the edge of the shell. For some individuals, a clean measurement was not possible. Either because the oysters were broken due to handling or because the line was not clearly visible. However, at least 7 or more oysters were measured per treatment. The survival rate for all concentrations and the control was 100%.
