Fusarium species of the Gibberella fujikuroi species complex cause serious diseases on different crops such as rice, wheat and maize. An important group of plant pathogens is the Gibberella fujikuroi species complex (GFC) of closely related Fusarium species which are associated with specific hosts; F. verticillioides and F. proliferatum are particularly associated with maize where they can cause serious ear-, root-, and stalk rot diseases. Two other closely related species of the GFC, F. mangiferae and F. fujikuroi, which share about 90Prozent sequence identity with F. verticillioides, are pathogens on mango and rice, respectively. All of these species produce a broad spectrum of secondary metabolites such as phytohormones (gibberellins, auxins, and cytokinins), and harmful mycotoxins, such as fumonisin, fusarin C, or fusaric acid in large quantities. However, the spectrum of those mycotoxins might differ between closely related species suggesting that secondary metabolites might be determinants for host specificity. In this project, we will study the potential impact of secondary metabolites (i.e. phytohormones and certain mycotoxins) and some other species-specific factors (e.g. species-specific transcription factors) on host specificity. The recently sequenced genomes of F. mangiferae and F. fujikuroi by our groups and the planned sequencing of F. proliferatum will help to identify such determinants by genetic manipulation of the appropriate metabolic pathway(s).

Eine Brennstoffzelle als Primärenergiequelle mit einem Doppelschichtkondensator (Supercap) als Zwischenspeicher zu kombinieren ist ein vielversprechender Ansatz für zukünftige Elektrofahrzeuge. In Kooperation mit einem Fahrzeughersteller wurden verschiedene Strategien für ein Energiemanagement für die Kombination einer Brennstoffzelle mit einem Doppelschichtkondensatormodul entworfen und verglichen. Basierend auf der aktuellen Geschwindigkeit und Beschleunigung werden verschiedene Fahrzeugzustände bezüglich kinetischer Energie und Leistungsbedarf unterschieden. In Abhängigkeit von der verfügbaren Leistung von Supercaps und Brennstoffzelle wird eine optimale Leistungsaufteilung zwischen den beiden Energiequellen ermittelt. In Bremsphasen wird durch Rekuperation Energie zurückgewonnen und in den Supercaps gespeichert. Wenn die Supercaps vollgeladen sind oder ihre maximale Ladeleistung erreicht haben, übernehmen mechanische Bremsen die übrige Ladeleistung. Da diese Situation zu einem Energieverlust führt, sollte sie möglichst vermieden werden. Um immer die notwendige Beschleunigungsleistung und gleichzeitig auch ein Maximum an Rekuperation zu garantieren, wird der Ladezustand der Supercaps kontinuierlich und dynamisch an die kinetische Energie des Fahrzeugs angepasst. Verschiedene Strategien wurden in Matlab/Simulink mit einem Stateflow-Chart zur Abbildung der Zustände implementiert. Die verfügbare Supercapleistung wird mit Hilfe eines impedanzbasierten Modells für Supercaps berechnet. Mit diesen Strategiemodellen können die Leistungsfähigkeit der verschiedenen Strategien verglichen und die Einflüsse von Parametern untersucht werden. Ziel eines Energiemanagements ist es, den Wasserstoffverbrauch zu minimieren und die notwendige Leistung zu jeder Zeit sicherzustellen. Bei der Bewertung der Strategien wird der Wasserstoffverbrauch, die verlorene Bremsenergie und eine mögliche Geschwindigkeitsreduzierung verglichen. Mit einer optimalen Strategie können bis zu 23 Prozent Wasserstoff während eines definierten Fahrprofils gespart werden.
Atmospheric CO2 concentrations present a repetitive pattern of gradual decline and rapid increase during the last climate cycles, closely related to temperature and sea level change. During the Last Glacial Maximum (LGM; 23-19 kyr BP), when sea level was ca. 120 m below present, the ocean must have stored additionally about 750 Gt carbon. There is consensus that the Southern Ocean represents a key area governing past and present CO2 change. The latter is not only of high scientific but also of socio-economic and political concern since the Southern Ocean provides the potential for an efficient sink of anthropogenic carbon. However, the sensitivity of this carbon sink to climate-change induced reorganizations in wind patterns, ocean circulation, stratification, sea ice extent and biological production remains under debate. Models were not yet able to reproduce the necessary mechanisms involved, potentially due to a lack of the dynamic representation/resolution of atmospheric and oceanic circulation as well as missing carbon cycling. Data on past Southern Ocean hydrography and productivity are mainly from the Atlantic sector, thus do not adequately document conditions in the Pacific sector. This sector is not only the largest part of the Southern Ocean, but it also represents the main drainage area of the marine-based West Antarctic Ice Sheet (WAIS). In the proposed study we aim to generate paleo-data sets with a newly established proxy method from sediment core transects across the Pacific Southern Ocean. This will enhance the baselines for the understanding and modeling of the Southern Ocean's role in carbon cyling, i.e. ocean/atmosphere CO2 exchange and carbon sequestration. It will also allow insight into the response of the WAIS to past warmer than present conditions. Paired isotope measurements (oxygen, silicon) will be made on purified diatoms and radiolarians to describe glacial/interglacial contrasts in physical and nutrient properties at surface and subsurface water depth. This will be used to test (i) the impact of yet unconsidered dust-borne micronutrient deposition on the glacial South Pacific on shifts of primary productivity, Si-uptake rates and carbon export, (ii) the 'silicic-acid leakage' hypothesis (SALH) and (iii) the formation and extent of surface water stratification. Diatom and radiolarian oxygen isotopes will provide information on the timing of surface ocean salinity anomalies resulting from WAIS melt water. Climate model simulations using a complex coupled atmosphere ocean general circulation model (AOGCM) in combination with a sophisticated ocean biogeochemical model including Si-isotopes will be used for comparison with the paleo records. The analysis will cover spatial as well as temporal variability patterns of Southern Ocean hydrography, nutrient cycling and air-sea CO2-exchange. Various species of pest insects cause substantial damage to agriculture every year, or transmit deadly diseases to animals and humans. A successful strategy to control pest insect populations is based on the Sterile Insect Technique (SIT), which uses the release of mass-reared, radiation sterilized male insects to cause infertile matings and thus reduce the pest population level. However, irradiation is not applicable to every insect species. Thus, new strategies based on genetic modifications of pest insects have been developed or are currently under investigation.The goal of the proposed research is to improve the development and ecological safety of genetically engineered (GE) insects created for enhanced biological control programs, including the SIT and new strategies based on conditional lethality. A major concern for GE insect release programs is transgene stability, and maintenance of their consistent expression. Transgene loss or intra-genomic movement could result in loss of strain attributes, and may ultimately lead to interspecies movement resulting in ecological risks. To address potential transgene instability, a new transposon vector that allows post-integration immobilization will be tested in the Mediterranean, Mexican and Oriental fruit fly tephritid pest species. In addition, the system will be established in the mosquito species Aedes and Anopheles - carriers of dengue and malaria.Random genomic insertion is also problematic for GE strain development due to genomic position effects that suppress transgene expression, and insertional mutations that negatively affect host fitness and viability. Diminished transgene expression could result in the unintended survival of conditional lethal individuals, or the inability to identify them. To target transgene vectors to defined genomic insertion sites having minimal negative effects on gene expression and host fitness, a recombinase-mediated cassette exchange (RMCE) strategy will be developed that. RMCE will also allow for stabilization of the target site, will be tested in tephritid and mosquito species, and will aid to the development of stabilized target-site strains for conditional lethal biocontrol. This will include a molecular and organismal evaluation of an RNAi-based lethality approach. Lethality based on an RNAi mechanism in the proposed insects would increase the species specificity and having multiple targets for lethality versus one target in existing systems. By seeking to improve transgene expressivity and stabilization of transposon-based vector systems, this proposal specifically addresses issues related to new GE insects by reducing their unintended spread after field release, and by limiting the possibilities for transgene introgression.
The formation of biogeochemical interfaces in soils is controlled, among other factors, by the type of particle surfaces present and the assemblage of organic matter and mineral particles. Therefore, the formation and maturation of interfaces is studied with artificial soils which are produced in long-term biogeochemical laboratory incubation experiments (3, 6, 12, 18 months. Clay minerals, iron oxides and charcoal are used as major model components controlling the formation of interfaces because they exhibit high surface area and microporosity. Soil interface characteristics have been analyzed by several groups involved in the priority program for formation of organo-mineral interfaces, sorptive and thermal interface properties, microbial community structure and function. Already after 6 months of incubation, the artificial soils exhibited different properties in relation to their composition. A unique dataset evolves on the development and the dynamics of interfaces in soil in the different projects contributing to this experiment. An integrated analysis based on a conceptual model and multivariate statistics will help to understand overall processes leading to the biogeochemical properties of interfaces in soil, that are the basis for their functions in ecosystems. Therefore, we propose to establish an integrative project for the evaluation of data obtained and for publication of synergistic work, which will bring the results to a higher level of understanding.
