The Swiss contribution to the ICDP Lake Van Drilling Project: Linking modern seismic and biogeochemical signatures to 500,000 years of environmental history

This SNF proposal seeks funding for the continuation of the Swiss contribution to the Lake Van Drilling Project executed by the International Continental Scientific Drilling Program (ICDP). Among other previous Swiss ICDP engagements, the Lake Van Drilling project was pivotal in triggering the newly established SNF-supported Swiss membership in ICDP. Further, the SNF Swiss contribution is a central building block of the entire ICDP PaleoVan initiative. Lake Van is the fourth-largest terminal lake in the world, extending 130 km WSW-ENE 1674 m above sea level on a high plateau in eastern Anatolia, Turkey. The lake is surrounded by active volcanoes within a tectonically active area and it is known to accumulate fluids emanating from the Earths mantle. The partly annually-laminated sedimentary record down to 220 m depth recovered from Lake Van during the ICDP PaleoVan drilling operations in 2010 has been shown to be an excellent palaeoclimate and palaeoenvironment archive. The continuous, high-resolution continental sequence, which covers several glacial-interglacial cycles (greater than 500 kyr), represents a unique possibility to investigate in detail the climatic, environmental, and volcanic changes that occurred in the Near East, the cradle of human civilization, during much of the Quaternary Period. Furthermore, the sediments contain an invaluable record of past earthquake activities, allowing the construction of a catalogue of prehistoric earthquakes and making it possible to study fluid transport in the continental crust that was triggered by seismic events. In this context, the societal vulnerability of the area to seismic hazards was dramatically documented by the occurrence of the devastating earthquake of magnitude 7.2 close to the city of Van on 23 October 2011 (hereafter referred to as the VE11 earthquake). This unfortunate and tragic event offers a unique opportunity to calibrate the past seismic events recorded in the sediments of Lake Van and the related emission of fluids from the solid earth to a modern seismic analogue. Sediment and fluid transport triggered by this major seismic event need to be quantified in order to calibrate the sedimentological record, which is targeted by the follow-up field campaign proposed within this project extension. The continuation of the Swiss initiative, embedded in the overarching ICDP drilling project on Lake Van, encompasses all the 5 initial research modules (A-E) of the ongoing SNF project (200021-124981). Within this proposal extension, the extended modules (A*-E*) will focus on key issues and new developments that expand the initial topics, with a special emphasis on the recent major earthquake VE11 and its biogeochemical and sedimentological implications. At the same time, this extension will also allow the results that have already been acquired to be further analysed and written up for publication by the project team. A large number of publications is foreseen. (...)

Secure production of a vaccine against Leishmaniasis: Expression of a Leish-111-antigen, precisely located in the plastome

Production of pharmaceutics in plants is up to 50 times more cost effective than in fermenters through micro organisms. Plant derived drugs are an interesting alternative for developing countries where people cannot afford sufficient medical treatment. Therefore it makes sense to use this approach for synthesis of vaccines. Vaccines can be delivered as antigens either for peripheral or oral immunization. Transformation of plants with genes carrying selected antigens of the respective pathogen allows producing immunogenic proteins on the field with high yields. However, transformation of the nucleus leads to transgenic pollen which is spread to the environment. Our solution to this ecological problem consists in the alternative to transform the chloroplasts. In contrast to the nucleus tobacco plastids are not contained in the pollen, and thus can not be dispersed in the vicinity. Our goal is the production of an antigen vaccine against Leishmaniasis. According to the World Health Organization Leishmaniasis is one of the most serious, endemic parasitic infections afflicting the poor and disadvantaged in many countries of the world, particularly in North Africa, most of Asia, parts of the Middle East and much of South America. 12 million cases worldwide and an estimated 350 million people at risk for acquiring infection are assumed. In order to produce the antigenic Leish-111 epitope a chloroplast transformation vector will be constructed, which will contain a synthetic expression cassette for increased transcription conferred by two different promoters and synthetic ribosomal binding sites. Further the Leish-111 gene will be N-terminally fused to the sequence codons of a peptide which confers an increased translation efficiency. On transcriptional level this construct is followed by a selection cassette containing the aadA gene. Following selection on a medium containing spectinomycin positive transformants will be identified by PCR and Southern analysis. Further Western blot analysis will prove successful expression of the Leish-111 protein. Novel transformants are then tested on correct folding of the fusion protein. Depending on the upcoming results the next step will be to proof the immunogenicity and protectivity. The further work is aimed to set up an inducible antigen expression system, which is regulated through chemical induction.

Schwerpunktprogramm (SPP) 1006: Bereich Infrastruktur - Internationales Kontinentales Bohrprogramm, Sub project: Bathymetric Survey of Lake Van (BathyVan), Turkey - finding answers to objectives not directly addressed by ICDP drilling at Lake Van

As Lake Van (Eastern Anatolia) can act as a key site for the investigation of the Quaternary climatic evolution of the Near East, an ICDP drilling campaign was successfully conducted in summer 2010. Preliminarily first core analyses show that the drilled sediments contain 500,000 years of paleo-environmental and volcanic/geodynamic history. Due to financial limitations, only 2 out of 5 primary proposed sites were drilled leaving some of the key questions of the drilling proposal nearly untouched including lake-level history, tectonics, and the interactions between man and environment. First results from the drilling campaign, such as tephra deposits formed underwater and contrasting pore water profiles from the drilled sites, bring up new questions, including the occurrence of sub-aquatic volcanic activity and the hydro-geological evolution of individual basins. We propose to address these questions (tectonics, sub-aquatic volcanology, geo-hazards, lake level history, seepage, and sub-aquatic archeology) by collecting a high resolution bathymetric data set of Lake Van. The work should be complemented by seismic data acquisition in selected areas and gravity coring of specific features, such as historic slides and sediments around faults and volcanic edifices. The new data in combination with the drilling and other already available data will allow addressing several questions of the original ICDP proposal, hence, contributing significantly to the overall success of the entire drilling campaign.
