Molecular and biochemical characterization of lipoxygenase candidate genes involved in the biosynthesis of flavour-related volatiles in cultivated apple (Malus x domestica)

Lipoxygenases (LOX) catalyze the addition of molecular oxygen to polyunsaturated fatty acids to produce their respective hydroperoxides and have many different putative physiological functions in higher plants. The LOX reaction provides the substrates, e.g. for the formation of aroma-active C6-aldehydes as well as hexyl and hexenyl esters that decisively shape the aroma pattern of ripe and stored apple fruits. Therefore, the focus of the project will be on the characterization of the LOX gene family in apple to investigate for the first time crucial steps of the aroma formation in fruits of the cultivated apple with an interdisciplinary approach using molecular genetic and biochemical methods. Preliminary data obtained from QTL analyses and candidate gene mapping approaches have confirmed that lipoxygenase (LOX) is an important gene which determines aroma profiles of apple genotypes. Available Malus databases will be screened for LOX nucleotide sequences. LOX genes belonging to different subclasses of the gene family will be cloned by PCR-based strategies and genetically mapped in the Malus genome. Temporal and spatial expression patterns will be determined and candidate genes functionally characterized by heterologous expression systems (E. coli, Nicotiana, Fragaria). The data will finally be used to develop functional markers to support molecular breeding and to enable a more efficient documentation of biodiversity of aroma patterns in Malus gene bank accessions, apple varieties and breeding material lifting the selection of suitable crossing partners to a new efficiency level.

One-year high-resolution time series of organic trace substances in a Baltic Sea urban estuary and adjacent areas in Germany (2022–2023)

A highly temporally and spatially resolved measurement campaign was conducted over a one-year period to analyze water samples for a total of 35 pharmaceuticals, pesticides and UV-filters. Between spring 2022 and spring 2023, surface water samples were collected twice a week at 14 sites at the Warnow estuary and its adjacent areas in Rostock, Germany, as part of the OTC-Genomics sampling campaign. The sampling area included one upstream freshwater site before the river enters the estuary, seven sampling sites along the estuary - surrounded by the urban area of Rostock - and six sites along the Baltic coastline. Sampling was conducted every Monday and Thursday, always three hours after sunrise. After enrichment with solid phase extraction, 1307 samples were analyzed using liquid chromatography–tandem mass spectrometry. Additional to the dataset included here, the same samples were analyzed for prokaryotic and eukaryotic community composition (16S and 18S rRNA gene amplicon sequencing), cell abundances (flow cytometry), and physico-chemical environmental parameters.

Entwicklung einer Methode zur Aufarbeitung gebrauchter und minderwertiger nativer Fette und Öle zu Treibstoff für Dieselmaschinen

Gebrauchte oder minderwertige native Fette und Öle sind eine interessante Energiequelle für Dieselmaschinen, die sich durch eine ausgezeichnete Ökobilanz auszeichnen und nicht in Konkurrenz zu Nahrungs- oder Futtermitteln stehen. Dem Einsatz in Dieselmschinen stehen der i.d.R. hohe Gehalt an Schlackebildnern (Ca, Mg, Na, K, P) und an freien Fettsäuren entgegen. Ziel des Vorhabens ist es, ein Verfahren zu entwickeln, mit dem die o.g. Rohstoffe so aufzuarbeiten sind, dass sie ohne weiteres in Dieselmaschinen eingesetzt werden können. Dazu wurde der Rohstoff einer sauer katalysierten Veresterung mit biogenem Ethanol unterworfen, mit dem die Gehalte sowohl an freien Fettsäuren, als auch an den genannten Schlackebildnern soweit gesenkt werden konnten, dass die Maßgaben der DIN-VN 51 605 erfüllt werden. Abgesehen davon, dass die so gewonnen Treibstoffe aus rein biogenen Rohstoffen bestehen, weisen sie Stockpunkte von teilweise unter -20 Grad Celsius auf.

Ein neuer Selenocarbamat Ester wurde dargestellt und dessen Reaktion mit verschiedenen Gold(I)-Komplexen wurde untersucht. Die erhaltenen Substanzen wurden spektroskopisch und strukturell charakterisiert.

Darstellung und Optimierung von molekularen Wirten für Phosphat und ähnliche Oxoanionen

Die molekulare Erkennung von Phosphat und seinen Estern durch künstliche Wirtstrukturen (sog. abiotische Rezeptoren) ist nicht befriedigend gelöst. Dabei fänden erfolgreiche Phosphatrezeptoren unmittelbar Anwendung in ionenselektiven Elektroden z.B. für die Umweltanalytik bei der Überwachung von Gewässern. Im Zuge von Vorprojekten haben wir die Grundlagen der Phosphatbindung in wasserähnlichen Lösungsmitteln erkundet und konnten Wirte mit sehr aussichtsreichen Eigenschaften darstellen. Das vorliegende Projekt soll diese Wirte fortentwickeln und das zugrundeliegende Konzept überprüfen. In Ergänzung des bisherigen Ansatzes soll ein neuer Rezeptorbautyp hergestellt und erprobt werden, der die bekannten Prinzipien der Phosphatbindung berücksichtigt, aber vermutlich wesentlich einfacher zugänglich ist. Die kalorimetrische Vermessung der thermodynamischen Bindungsparameter liefert die Leitlinie für die Auswahl geeigneter Rezeptoren für die geplanten ionenselektiven Anwendungen.

Les principaux polluants etudies sont: les polychlorobiphenyles (PCB), les hydrocarbures aromatiques polycycliques (PAH), les tensio-actifs (nonyl-phenol et LAS), les organoetains, les esters phosphoriques et les chloro- et nitro-phenols.
