The basidiomycete Armillaria mellea s.l. is one of the most important root rot pathogens of forest trees and comprises several species. The aim of the project is to identify the taxa occurring inSwitzerland and to understand their ecological behaviour. Root, butt and stem rots caused by different fungi are important tree diseases responsible for significant economic losses. Armillaria spp. occur world-wide and are important components of many natural and managed forest ecosystems. Armillaria spp. are known saprothrophs as well as primary and secondary pathogens causing root and butt rot on a large number of woody plants, including forest and orchard trees as well as grape vine and ornamentals. The identification of several Armillaria species in Europe warrants research in the biology and ecology of the different species. We propose to study A. cepistipes for the following reasons. First, A. cepistipes is dominating the rhizomorph populations in most forest types in Switzerland. This widespread occurrence contrasts with the current knowledge about A. cepistipes, which is very limited. Second, because the pathogenicity of A. cepistipes is considered low this fungus has the potential for using as an antagonist to control stump colonising pathogenic fungi, such as A. ostoyae and Heterobasidion annosum. This project aims to provide a better understanding of the ecology of A. cepistipes in mountainous Norway spruce (Picea abies) forests. Special emphasis will be given to interactions of A. cepistipes with A. ostoyae, which is a very common facultative pathogen and which often co-occurs with A. cepistipes. The populations of A. cepistipes and A. ostoyae will be investigated in mountainous spruce forests were both species coexist. The fungi will be sampled from the soil, from stumps and dead wood, and from the root system of infected trees to determine the main niches occupied by the two species. Somatic incompatibility will be used to characterise the populations of each species. The knowledge of the spatial distribution of individual genets will allow us to gain insights into the mode of competition and the mode of spreading. Inoculation experiments will be used to determine the variation in virulence expression of A. cepistipes towards Norway spruce and to investigate its interactions with A. ostoyae.
The aim of P2 within the Research Unit 'The Forgotten Part of Carbon Cycling: Organic Matter Storage and Turnover in Subsoils (SUBSOM)' is to contribute to the understanding of the different sources and stabilization processes of subsoil organic matter. This will be achieved by the analysis of the soil organic matter composition in topsoil versus subsoil by 13C NMR spectroscopy in bulk soils as well as organo-mineral associations. This will be done on a number of soil profiles differing in parent material and mineralogy and therefore also in the relevance of organo-mineral associations for subsoil C stabilization. In addition, a specific sampling approach will allow to differentiate three zones associated with the dominating effect of (1) leaching of DOC (the 'bulk soil' between trees), (2) root litter decomposition (the 'root-affected zone'), and (3) direct rhizodeposition of root exudates (the 'rhizosphere' sensu strictu). The contribution of above-ground versus below-ground litter is differentiated by the analysis of cutin and suberin biomarkers. Organic matter derived from microbial sources will be identified by the microbial signature of polysaccharides in the subsoil through the analysis of neutral sugars and amino sugars. Organo-mineral associations will be further characterized by N2-BET analyses to delineate the coverage of the mineral phase with organic matter. With these analyses and our specific analytical expertise at the submicron scale (nanoSIMS) we will participate in selected joint experiments of the research unit.
Um aktuelle Umwelt- und Klimaveränderungen in einem längeren zeitlichen Kontext bewerten zu können, insbesondere anthropogene und natürliche Einflüsse auf den atmosphärischen Aerosoleintrag in die Atmosphäre zu verstehen, werden Informationen über die Zusammensetzung der Atmosphäre in der Vergangenheit benötigt. Eisbohrkerne aus den Polarregionen oder Gletschern sind wertvolle Umwelt- und Klimaarchive, da sie unter anderem das mit dem Schnellfall deponierte Aerosol enthalten. Daher kann die Analyse von partikel-gebundenen Spurenstoffen in Eisbohrkernen stoffliche Informationen über zurückliegende Umwelt- und Klimabedingungen liefern. Bis heute konzentrieren sich diese Bemühungen auf anorganische Substanzen und nur wenige organische Analyten. Ein Großteil der in diesen Bohrkernen enthaltenen Informationen geht dadurch verloren. Dies charakterisiert das Hauptziel des Vorhabens, durch Erarbeitung organischer spurenanalytischer Methoden basierend auf LC-HRMS (Flüssigchromatographie in Kombination mit hochauflösender Massenspektrometrie) eine ausgewählte Palette von Markersubstanzen zu quantifizieren. Zielmoleküle sind insbesondere neue Marker für biogene sekundäre organische Aerosole (biogenic SOA) und Biomasseverbrennungs-Marker. Die Auswahl dieser Verbindungen basiert einerseits auf dem zu erwartenden Informationsgewinn über die Quellen und deren Zusammensetzung (terrestrische Vegetation/Waldbrände), andererseits auf der atmosphärischen Lebensdauer der Marker, da nur langlebige Marker weit entfernt liegende Regionen erreichen können. Zusätzlich zur Analyse dieser Zielanalyten sollen auch ââ‚ Ìnon-target screening-Methoden zum Einsatz kommen. In enger Zusammenarbeit mit einem etablierten Eisbohrkernlabor am PSI in der Schweiz werden die entwickelten Analysetechniken auf einen Eisbohrkern aus dem Belukha-Gletscher im Sibirischen Altai Gebirge angewendet.
It is well established that reduced supply of fresh organic matter, interactions of organic matter with mineral phases and spatial inaccessibility affect C stocks in subsoils. However, quantitative information required for a better understanding of the contribution of each of the different processes to C sequestration in subsoils and for improvements of subsoil C models is scarce. The same is true for the main controlling factors of the decomposition rates of soil organic matter in subsoils. Moreover, information on spatial variabilities of different properties in the subsoil is rare. The few studies available which couple near and middle infrared spectroscopy (NIRS/MIRS) with geostatistical approaches indicate a potential for the creation of spatial maps which may show hot spots with increased biological activities in the soil profile and their effects on the distribution of C contents. Objectives are (i) to determine the mean residence time of subsoil C in different fractions by applying fractionation procedures in combination with 14C measurements; (ii) to study the effects of water content, input of 13C-labelled roots and dissolved organic matter and spatial inaccessibility on C turnover in an automatic microcosm system; (iii) to determine general soil properties and soil biological and chemical characteristics using NIRS and MIRS, and (iv) to extrapolate the measured and estimated soil properties to the vertical profiles by using different spatial interpolation techniques. For the NIRS/MIRS applications, sample pretreatment (air-dried vs. freeze-dried samples) and calibration procedures (a modified partial least square (MPLS) approach vs. a genetic algorithm coupled with MPLS or PLS) will be optimized. We hypothesize that the combined application of chemical fractionation in combination with 14C measurements and the results of the incubation experiments will give the pool sizes of passive, intermediate, labile and very labile C and N and the mean residence times of labile and very labile C and N. These results will make it possible to initialize the new quantitative model to be developed by subproject PC. Additionally, we hypothesize that the sample pretreatment 'freeze-drying' will be more useful for the estimation of soil biological characteristics than air-drying. The GA-MPLS and GA-PLS approaches are expected to give better estimates of the soil characteristics than the MPLS and PLS approaches. The spatial maps for the different subsoil characteristics in combination with the spatial maps of temperature and water contents will presumably enable us to explain the spatial heterogeneity of C contents.
*Der Gesundheitszustand der Bäume im Schweizer Wald wird seit 1985 mit der Sanasilva-Inventur repräsentativ erfasst. Die wichtigsten Merkmale sind die Kronenverlichtung und die Sterberate. Das systematische Probeflächen-Netz der Inventur ist im Laufe der Zeit ausgedünnt worden. In der Periode von 1985 bis 1992 wurden rund 8000 Bäume auf 700 Flächen im 4x4 km-Netz aufgenommen, 1993, 1994 und 1997 rund 4000 Bäume im 8x8 km-Netz und in den Jahren 1995, 1996 und 1998 bis 2002 rund 1100 Bäume im 16x16 km-Netz . Aufnahmemethode Alle drei Jahre (1997, 2000) wird die Sanasilva-Inventur auf dem 8x8-km Netz (ca. 170 Probeflächen ) durchgeführt. In den Jahren dazwischen findet die Inventur auf einem reduzierten 16x16-km Netz (49 Probeflächen) statt. Jede Fläche besteht aus zwei konzentrischen Kreisen. Der äussere Kreis hat ein Radius von 12.62 m (500 m2) und der innere ein Radius von 7.98 m (200 m2). Auf dem inneren Kreis werden alle Bäume mit einem Mindestdurchmesser in Brusthöhe von 12 cm und auf dem äusseren Kreis mit einem Mindestdurchmesser in Brusthöhe von 36 cm aufgenommen. In Nordrichtung wird zusätzlich in 30 m Entfernung eine identische Satellitenprobenfläche eingerichtet. Die Aufnahme findet in Juli und August statt. Eine Aufnahmegruppe besteht aus zwei Personen, von denen eine die Daten erhebt, und die andere die Daten eintippt. Die Daten werden mit dem Feldkomputer Paravant und der Software Tally erfasst. Die Aufgabenteilung wechselt zwischen Probeflächen. Auf dem 8x8-km Netz werden zusätzlich 10 Prozent der Flächen von einer unabhängigen zweiten Aufnahmegruppe zu Kontrollzwecken aufgenommen. Hauptmerkmale der Sanasilva-Inventur: Die Sanasilva-Inventur erfasst vor allem folgende Indikatoren des Baumzustandes: Die Kronenverlichtung wird beschrieben durch den Prozentanteil der Verlichtung einer Krone im Vergleich zu einem Baum gleichen Alters mit maximaler Belaubung/Benadelung an diesem Standort, den Anteil dieser Verlichtung, der nicht durch bekannte Ursachen erklärt werden kann, den Ort der Verlichtung, den Anteil und den Ort von unbelaubten/unbenadelten Ästen und Zweigen. Die Kronenverfärbung wird durch die Abweichung der mittleren Farbe (aufgenommen als Farbton, Reinheit und Helligkeit nach den Munsell Colour Charts) eines Baumes zu der für diese Baumart typischen Normalfarbe (Referenzfarbe) und durch das Vorhandensein, das Ausmass und den Ort der von der Referenzfarbe abweichenden Farben beschrieben. Der Zuwachs eines Baumes wird durch die zeitliche Veränderung der aufgenommen Baumgrössen beschrieben (Brusthöhendurchmesser, Höhe des Baumes, Kronenlänge und Kronenbreite). Weitere Merkmale sind die erkannten Ursachen der Kronenverlichtung, die Kronenkonkurrenz und das Vorkommen von Epiphyten, Mistel und Ranken in der Baumkrone.
Der Datensatz beinhaltet Informationen zur Geologie im Freistaat Sachsen. Der Datensatz des Kartenwerkes bildet die tertiären Lockergesteine und die an ihrer Basis anstehenden Festgesteine in mehreren, untereinanderfolgenden Horizontkarten im Maßstab 1 : 50.000 ab. Lückenhafte Angaben zu Stratigraphie und/oder Petrographie einzelner Flächen werden schrittweise vervollständigt. Eine einheitlich graue Signatur markiert die unvollständig attributierten Features. Der Datensatz ist nutzbar in einem Maßstab kleiner als 1 : 25.000. Das Kartenwerk ist eines von 7 geologischen Kartenwerken, welches für das Gebiet des Freistaates Sachsen in jeweils eigenen Kartendiensten INSPIRE-konform bereitgestellt wird.
Abfaelle aus polyolefinischen Materialien fallen einerseits in grossen Mengen in Form von Verpackungsmaterial oder Ein-Weg-Gebrauchsgegenstaenden beim Endverbraucher an. Andererseits werden auch bei der Herstellung von Polyolefinen, je nach Herstellungsverfahren und -bedingungen niedermolekulare und wachsartige Nebenprodukte erhalten, die nur zum geringen Teil Verwendung finden. Diese Abfaelle - sowohl die Nebenprodukte aus der Produktion als auch die Abfaelle aus dem Endverbrauch - werden zum groessten Teil verworfen und finden nur zum geringen Teil Anwendung, z.B. bei der Dampferzeugung in Kraftwerken oder Muellverbrennungsanlagen. Mit dem Forschungsprojekt soll daher geprueft werden, wie weit aus diesen Polyolefinabfaellen die Rohstoffe - Aethylen oder Propylen - oder andere Komponenten der chemischen Grundstoffproduktion - z.B. Acetylen - gewonnen werden koennen.Bei den entwickelten Verfahren wurden, im Gegensatz zu den mechanisch-thermischen Aufbereitungsverfahren, die Polyolefine einer partiellen Oxidation unterworfen. Bei dem Forschungsprojekt wurde zunaechst von ataktischem Polypropylen ausgegangen. Dies wurde aufgeschmolzen und in einem Injektionsbrenner zerstaeubt und anschliessend in einer Brennkammer mit Sauerstoff partiell oxidiert. Der Oxidationsvorgang wird dabei durch die Eigenschaften des Brennstoffnebels - Troepfchengroesse, Relativgeschwindigkeit Troepfchen/Gas- und durch die Menge des im Unterschuss eingesetzten Sauerstoffs beeinflusst. Hierdurch laesst sich die Produktverteilung bei der partiellen Oxidation, insbesondere die Konzentration an Olefinen und Acetylen, in relativ weiten Grenzen steuern.
Comprehension of belowground competition between plant species is a central part in understanding the complex interactions in intercropped agricultural systems, between crops and weeds as well as in natural ecosystems. So far, no simple and rapid method for species discrimination of roots in the soil exists. We will be developing a method for root discrimination of various species based on Fourier Transform Infrared (FTIR)-Attenuated Total Reflexion (ATR) Spectroscopy and expanding its application to the field. The absorbance patterns of FTIR-ATR spectra represent the chemical sample composition like an individual fingerprint. By means of multivariate methods, spectra will be grouped according to spectral and chemical similarity in order to achieve species discrimination. We will investigate pea and oat roots as well as maize and barnyard grass roots using various cultivars/proveniences grown in the greenhouse. Pea and oat are recommendable species for intercropping to achieve superior grain and protein yields in an environmentally sustainable manner. To evaluate the effects of intercropping on root distribution in the field, root segments will be measured directly at the soil profile wall using a mobile FTIR spectrometer. By extracting the main root compounds (lipids, proteins, carbohydrates) and recording their FTIR-ATR spectra as references, we will elucidate the chemical basis of species-specific differences.
