Schwerpunktprogramm (SPP) 1530: Flowering time control: from natural variation to crop improvement, Metabolit-Regulation von Cryptochrom 2 Aktivität und Blühzeitpunkt

Der UV-A/Blaulichtrezeptor Cryptochrom 2 (cry2) spielt eine zentrale Rolle bei der photoperiodischen Blühinduktion. Wichtige Komponenten der Signalleitung von cry2 sind mehrere nah verwandte bHLH Transkriptionsfaktoren (CIB1, CIB2, CIB4, CIB5), die Blaulicht-abhängig an cry2 binden, als Heterodimere an nicht-kanonische E-Box Motive in der Promotorregion des FLOWERING LOCUS T (FT) Gens binden und hierdurch die Expression dieses zentralen Blühgens induzieren. Weiterhin interagiert cry2 im Blaulicht mit SPA-Proteinen und inhibiert damit die Aktivität der E3 Ubiquitin Ligase CONSTITUTIVE PHOTOMORPHOGENESIS 1 (COP1), die maßgeblich am Abbau des FT-Transkriptionsaktivators CONSTANS (CO) beteiligt ist. Wir haben gezeigt, dass der FAD-Chromophor im signalaktiven Zustand von cry2 in der neutralen Semichinonform vorliegt und die Bildung dieser Form durch Metabolite wie ATP und NADPH verstärkt wird. Gerichte und strukturbasierte Mutagenese von CRY2 lieferte Hinweise darauf, welche Aminosäuren für die Metabolit-Kontrolle erforderlich sind. Diese Information soll genutzt werden, um die Rolle dieser Metabolite bei der photoperiodischen Blühinduktion aufzuklären. Geplant ist hierfür die Expression entsprechender cry2 Allele in planta und die Analyse der generierten Pflanzenlinien hinsichtlich Blühverhalten, FT-Expression und Interaktion mit down-stream Komponenten. Diese Untersuchungen sollen durch in vitro Studien mit rekombinanten Proteinen komplettiert werden. Im Gegensatz zum Wildtyp Allel von cry1 induziert die cry1L407F Mutante gesteigerte Expression von CO und FT sowie frühes Blühen im Kurztag. In vitro Analysen zeigten ein tryptisches Spaltmuster von cry1L407F im Dunkeln, welches dem von Wildtyp cry1 im Blaulicht entspricht. Das geplante Vorhaben soll klären, ob cry1L407F die gleichen Komponenten wie cry2 für die Induktion von FT nutzt. Hierfür sind Analysen der cry1L407F Mutante im cib1/cib2/cib5 Hintergrund sowie Interaktionsstudien mit cry2 Partnern geplant. Direkte Zusammenarbeiten ergeben sich mit den Vorhaben von Christian Jung (Blühkontrolle bei Beta vulgaris) und Markus Schmid (Regulation des Blühzeitpunkts durch Trehalose-6-P).

Natural variation of flowering time due to cis-regulatory evolution of FLOWERING LOCUS T and its orthologs and paralogs in Brassica napus

In many plant species, FLOWERING LOCUS T and related proteins are the mobile signal that communicates information on photoperiod from the leaves to the shoots, where the transition to flowering is realized. FT expression is tightly controlled at the transcriptional level so that it is restricted to leaves, occurs only in appropriate photoperiods, and integrates ambient temperature and developmental cues, as well as information on biotic and abiotic stress. We previously established that FT transcription in the model plant Arabidopsis thaliana requires proximal promoter cis-elements and a distal enhancer, both evolutionary conserved among Brassicacea species. In addition, FT transcription is blocked prior vernalization in biannual accessions and vernalization-dependency of FT is controlled through a CArG-box located in the first intron that binds the transcriptional repressor FLOWERING LOCUS C (FLC). Chromatin-mediated repression by the Polycomb Group (PcG) pathway is required for photoperiod-dependent FT regulation and participates in FT expression level modulation in response to other cues.In this project, I propose to explore the available sequence data from the 1001 genome project in Arabidopsis to evaluate how often changes in regulatory cis-elements at FT have occurred and how these translate into an adaptive value. Allele-specific FT expression pattern will be measured in F1 hybrids of different accessions in response to varying environmental conditions. FT alleles that show cis-regulatory variation will be further analyzed to pinpoint the causal regulatory changes and study their effect in more detail. The allotetrapolyploid species Brassica napus is a hybrid of two Brassiceae species belonging to the A- and C-type genome, which are in turn mesopolyploid due to a genome triplication that occurred ca. 10x106 years ago. We will determine allele-specific expression of FT paralogs from both genomes of a collection of B. napus accessions. The plants will be grown in the field in changing environmental conditions to maximize the chance to detect expression variation of the paralogs. We will compare the contribution of the founder genomes to the regulation of flowering time and asses variation in this contribution. A particular focus will be to study the impact of chromatin-mediated repression on allele selection in B. napus.

The main objective of BETTER is to implement an EO Big Data intermediate service layer devoted to harnessing the potential of the Copernicus and Sentinel European EO data directly from the needs of the users. BETTER aims to go beyond the implementation of generic Big Data tools and incorporate those tools with user experience, expertise and resources to deliver an integrated Big Data intermediate service layer. This layer will deliver customized solutions denominated Data Pipelines for large volume EO and non-EO datasets access, retrieval, processing, analysis and visualisation. The BETTER solutions will focus in addressing the full data lifecycle needs associated with EO Big Data to bring more downstream users to the EO market and maximise exploitation of the current and future Copernicus data and information services. BETTER developments will be driven by a large number of Big Data Challenges to be set forward by the users deeply involved in addressing the Key Societal Challenges. The World Food Programme, the European Union Satellite Centre and the Swiss Federal Institute of Technology - Zurich working in the areas of Food Security, Secure Societies and GeoHazards will be the challenge promoters. During the project each promoter will introduce 9 challenges, 3 in each project year, with an additional nine brought by the 'Extending the market' task, in a total of 36 challenges. The Data Pipelines will be deployed on top of a mature EO data and service support ecosystem which has been under consolidation from previous R&D activities. The ecosystem and its further development in the scope of BETTER rely on the experience and versatility of the consortium team responsible for service/tool development from DEIMOS and Terradue. This is complemented by Fraunhofer Institute's experience in Big Data systems, which brings to the consortium transversal knowledge extraction technologies and tools that will help bridge the current gap between the EO and ICT sectors.

ERA-IB 5: CO2Chem - Biologische Konversion von CO2 zur Plattform-Chemikalie 3-Hydroxypropansäure, Teilprojekt Uni Ulm

Die globale Erderwärmung und die Versorgung mit Treibstoffen und Basischemikalien aus fossilen Quellen erfordern die Entwicklung alternativer, nachhaltiger Synthesewege für solche Substanzen. Biotechnologische Verfahren bieten eine Lösung, allerdings nur mit Substanzen, die nicht mit der menschlichen Ernährung konkurrieren. Neben Lignocellulose kommen dafür nur Gase wie CO2 oder CO in Frage. Hier soll CO2 mit Hilfe rekombinanter autotropher acetogener Bakterien in die industriell wichtige Plattformchemikalie 3-Hydroxypropanoat umgewandelt werden. Das ermöglicht eine kostengünstige Herstellung dieser Substanz, für die es keine ökonomisch tragfähige Synthese aus Rohölprodukten gibt, und die Reduktion eines bedeutenden Treibhausgases. Das Gen für Pyruvat-Synthase aus Acetobacterium woodii wird durch PCR amplifiziert und in einen geeigneten Schaukelvektor subkloniert (pJIR750). Zur Expression wird der Promoter des Acetat-Kinase-Gens aus A. woodii verwendet. Dann wird das Gen für Lactat-Dehydrogenase amplifiziert und subkloniert. Als Wirt für dieses Gen dient z.B. Clostridium acetobutylicum. Das dritte Gen kodiert eine Coenzym A-Transferase (Katalyse der Reaktionen: Lactat in Lactyl-CoA und 3-Hydroxypropanoyl-CoA in 3-Hydroxypropanoat). Als Wirt dient Megasphaera elsdenii. Es folgen Amplifikation und Subklonierung. Das vierte Gen kodiert eine Lactyl-CoA-Hydratase (Katalyse der Reaktion: Lactyl-CoA in Acrylyl-CoA). Als Wirt dient M. elsdenii. Es folgen Amplifikation und Subklonierung. Das fünfte Gen kodiert eine Acrylyl-CoA-Hydratase (Katalyse der Reaktion: Acrylyl-CoA in 3-Hydroxypropanoyl-CoA). Als Wirt dient Chloroflexus aurantiacus. Es folgen Amplifikation und Subklonierung. Analoge Gene werden auch aus C. propionicum, C. homopropionicum und C. neopropionicum kloniert und analysiert, um möglicherweise Patente zu generieren. Zum Schluss erfolgen Optimierung von Operon-Struktur und -Expression sowie Produktionstest in Laborkulturen und Kleinfermentern.

FP1-BAP, Genetic Manipulation of the anaerobe, Zymomonas Mobilis for Fermentation of waste Starch

Objective: Zymomonas bacteria could offer considerable potential for improved alcohol fermentation in comparison with yeasts. An efficient expression vector is, however, an absolute requisite to open the possibility of genetically engineered zymomonas. The bacteria could then be made able to use for example renewable carbon substrates abundant in agriculture wastes. General Information: An efficient expression vector shall be constructed for zymomonas mobilis. For this, the promotor of the pyruvate decarboxylase gene will be isolated, sequenced and built into a broad host range plasmid. Genes for the degradation of amylose (from aspergillus awamorii) or xylose (from klebsiella) shall be placed under control of this strong promoter and expression will be tested in suitable escherichia coli strains. Afterwards, these recombinant plasmids shall be introduced into zymomonas mobilis cells and their expression and stability will be assayed. The long range aim is to study the recombinant strains for continuous ethanol production from agricultural waste substrates as starch and hemicellulose (xylose). Achievements: Zymomonas mobilis is a Gram negative anaerobe producing ethanol 5 to 6 times faster than yeast. It can use only glucose, fructose and sucrose as carbon sources. Therefore, applications of Z mobilis at the industrial level require extention of its spectrum of usable carbohydrates. Genetic improvement was approached through formal genetics and genetic engineering. Chemical mutagenesis, transposon mutagenesis, gene transfer by aided conjugation, construction of recombinant expression vectors and transfer into Z mobilis genes responsible for xylose metabolism were established and can now be applied to increase the substrate range of Z mobilis. Recombinant plasmids were constructed by subcloning fragments of native Z mobilis plasmids in known vectors. These were transferred into Z mobilis by aided conjugation and were stably inherited, especially after the loss of native plasmids. Subcloned Z mobilis plasmid sequences were used to construct new expression vectors, which could be transferred into Z mobilis and induce expression of the xylose catabolism genes xylA and xylB. However, the engineered strains could not ferment xylose. Transformation of Z mobilis by plasmid deoxyribonucleic acid (DNA) occurred at low frequencies only when sequences of incoming plasmid were integrated into the host's genome. Transposon mutagenesis was accomplished by conjugal transfer of transposon bearing plasmids from Pseudomonas, as well as Escherichia coli donors to Z mobilis. All Z mobilis strains tested were able to ferment extracts of various fruits, such as apples, oranges, peaches, watermelons, etc.

Stoffwechsel und Wachstum der Pflanze unter erhoehter Kohlendioxidkonzentration - TP: Beeinflussung der Photoassimilatverteilung und der pflanzlichen Genexpression durch erhoehte CO2-Konzentration in der Atmosphaere unter besonderer Beruecksichtigu..

Die bisherigen Ergebnisse zeigen, dass Pflanzen, die unter erhoehter atmosphaerischer (CO2) angepflanzt wurden, eine schnellere Entwicklung durchlaufen. Die bei Kartoffelpflanzen beobachtete vorzeitige Knolleninduktion ist auf eine Erhoehung des C/N-Verhaeltnisses zurueckzufuehren. Erhoehung der Lichtintensitaet oder suboptimale Stickstoffversorgung fuehren zu vergleichbaren Ergebnissen. In der derzeitigen Antragsphase sollen vornehmlich drei Schwerpunkte bearbeitet werden. In allen bisherigen Untersuchungen wurde ein Anstieg AGPaseS-spezifischer Transkripte unter Hoch- CO2 beobachtet. Dieser Anstieg ist unabhaengig von der Staerkeakkumulation aber abhaengig vom C-Metabolismus. Zur Identifizierung moeglicher Signalgeber fuer die CO2-modulierte AGPaseS-Expression sollen transgene Pflanzen ausgenutzt werden, in denen die Synthese einzelner Metabolite vermindert ist. Im zweiten Teil soll der Zusammenhang zwischen erhoehter (CO2), der (GABA) und der Expression der ACC-Oxidase untersucht werden. Zur Ueberpruefung der Hypothese, dass bei erhoehter (CO2) eine Akkumulation von Ethylen auftritt, soll die (ACC) mittels HPLC in Blattscheiben bestimmt werden. In begleitenden Experimenten sollen transgene Pflanzen, die eine ACC-Oxidase spezifische-antisense RNA exprimieren, unter erhoehter (CO2) angepflanzt werden und der Einfluss einer verminderten Ethylenbiosynthese auf die Blattalterung und die Genexpression studiert werden. Im dritten Teil soll die Hypothese: Erhoehung der atmosphaerischen (CO2) fuehrt zu einer beschleunigten Seneszenz, geprueft werden. Hierzu stehen uns Pflanzen zur Verfuegung, die das GUS-Reportergen unter Kontrolle eines Seneszenz-spezifischen Promotors (SAG12; Gan und Amasino 1995) exprimieren, d.h. die Hoehe der GUS-Aktivitaet kann als Indikator fuer den Seneszenszustand des untersuchten Gewebes eingesetzt werden. Eine zweite Gruppe transgener Pflanzen exprimiert ein bakterielles Isopentenyltransferase (ipt) Gen unter Kontrolle des SAG12 Promotors. Expression des ipt Gens fuehrt zur Bildung von Cytokinin und damit zu einer Aufhebung der Seneszenz. Durch die Wahl des Promotors findet eine Autoregulation der Cytokininbildung statt, sodass negative Effekte zu hoher Phytohormonkonzentrationen nicht auftreten. Untersuchung dieser Pflanzen unter Hoch- CO2 sollte die einmalige Chance bieten Seneszenz- von CO2-induzierten Ereignissen zu unterscheiden. Neben den drei Hauptgebieten soll der Einfluss der Stickstoffversorgung auf CO2-vermittelte Seneszenzerscheinungen und die Auswirkung erhoehter (CO2) auf Resistenzeigenschaften von Tabakpflanzen untersucht werden.

Demonstration von Technologien zur Behandlung neuer Schadstoffe in der Wasser- und Abwasserreinigung (DEMEAU)

Das EU finanzierte Projekt DEMEAU ist ein dreijähriges Demonstrationsprojekt für vielversprechende Technologien zum Nachweis und zur Eliminierung von organischen Spurenstoffen im Wasserkreislauf. DEMEAU führt Ökobilanzen und Umweltbelastungsstudien für vier Technologiegruppen durch und fördert deren Anwendung. Die Technologien sind: künstliche Grundwasseranreicherung sowie Hybridlösungen von Keramikmembran­filtration und fortentwickelten Oxidationstechniken sowie Bioassays. Das DEMEAU Konsortium besteht aus 17 Mitgliedern aus fünf EU-Ländern und umfasst Wasserversorgungs­unternehmen mit deren Unterstützung die Anwendung der vielversprechenden Technologien demonstriert wird. Ziel des Projektes ist es, die Eignung und Wirtschaftlichkeit der innovativen Methoden und Technologien weiterzuentwickeln und im technischen Betrieb zu untersuchen.
