Evidence is compelling for a positive correlation between urbanisation and increment of allergic sensitisation and diseases. The reason for this association is not clear to date. Some data point to a pro-allergic effect of anthropogenic factors on susceptible individuals. Data analysing the impact of environmental - natural and anthropogenic - factors on the allergenicity of allergen carriers such as pollen grains are scarce, and if applicable only taken from in vitro experimental designs. This study will analyse one of the most common allergy inducers in northern Europe - the birch pollen. Under natural exposure conditions, birch pollen will be analysed with respect to their allergenicity. Within an interdisciplinary research team this study will evaluate the effect of natural (e.g. soil, climate, genetic background) and anthropogenic (e.g. traffic pollutants) factors on birch pollen in a holistic approach including analysis of allergen bioavailability, release of pollen associated lipid mediators from birch pollen grains, in vitro immunostimulatory activity and in vivo allergenic potential. These data collected in the time course of three years will significantly add to our understanding how urbanisation and climate change influence the allergenicity of birch pollen and will help us in the future to set up primary prevention studies.
In dem Vorhaben wird untersucht, wie wirksam die absorbierte Lichtenergie in Biomasse konvertiert wird. Vergleichend werden Grünalgen und Diatomeen unter verschiedenen Licht- und Nährstoffbedingungen studiert. Auf diese Weise können die metabolischen Kosten unter Nährstoffmangel oder anderen produktivitätsbegrenzenden Bedingungen studiert werden. So wird auch die Säureanpassung ausgewählter Phytoplankter untersucht, um die Biomassebildung in extrem sauren Tagebaurestseen auf physiologischer Ebene zu verstehen. Es konnte gezeigt werden, dass unter Stickstoffmangel die Überführung anorganischen Kohlenstoffs in Biomassebildung durch eine Veränderung der makromolekularen Zusammensetzung der Zellen ähnlicher Effizienz stattfindet, wie unter optimaler Stickstoffversorgung. Dies führt zu einer ökologisch bedeutsamen Teilentkopplung des C und N Kreislaufs im Ökosystem. Ähnliches beobachtet man auch bei der Anpassung von Phytoplanktonalgen an extrem saure Bedingungen wie man sie in sauren Tagebaurestseen vorfindet.
Almond in California represents an agroecosystem pollinated solely by a single species, the European honey bee, a species that is becoming increasingly difficult and expensive to manage due to substantial, unpredictable mortality. Therefore, sustainable and high output production require a more integrated approach that diversifies sources of pollination. For this purpose, detailed data of our understanding how diversity can stabilize pollination are required. The project will identify alternative wild pollinator species and collect high quality data contributing to our understanding of how diversity (pollen and insects) can bolster honey bee pollination during stable and unstable climatic conditions. The research will be carried out on almond orchards in Northern California known to be either pollinator species rich (up to 30 species) or depauperate (honey bees only). The replicated extremes in pollinator diversity represent a unique opportunity to study the effects of diversity on pollination in real agroecosystems combined with laboratory and glasshouse experiments. The overall goal is to provide basic research that is essential for our general understanding of how insect diversity can affect high-quality pollination under land use and climate change.
Soil structure determines a large part of the spatial heterogeneity in water storage and fluxes from the plot to the hillslope scale. In recent decades important progress in hydrological research has been achieved by including soil structure in hydrological models. One of the main problems herein remains the difficulty of measuring soil structure and quantifying its influence on hydrological processes. As soil structure is very often of biogenic origin (macropores), the main objective of this project is to use the influence of bioactivity and resulting soil structures to describe and support modelling of hydrological processes at different scales. Therefore, local scale bioactivity will be linked to local infiltration patterns under varying catchment conditions. At hillslope scale, the spatial distribution of bioactivity patterns will be linked to connectivity of subsurface structures to explain subsurface stormflow generation. Then we will apply species distribution modelling of key organisms in order to extrapolate the gained knowledge to the catchment scale. As on one hand, bioactivity influences the hydrological processes, but on the other hand the species distribution also depends on soil moisture contents, including the feedbacks between bioactivity and soil hydrology is pivotal for getting reliable predictions of catchment scale hydrological behavior under land use change and climate change.
Sekundäre Partikelneubildung ist eine Hauptquelle für atmosphärische Partikel mit wichtigen Folgen für das Klima und die menschliche Gesundheit. Dieses Vorhaben untersucht die Rolle von Luft Ionen bei der sekundären Partikelneubildung in Flussreaktor- und Aerosolkammer-Experimenten unter kontrollierten Laborbedingungen. Trotz beträchtlicher Fortschritte in der Messtechnik zur Untersuchung der atmosphärischen Nukleation und des Partikelwachstums bestehen weiterhin Verständnislücken hinsichtlich der grundlegenden physikalischen und chemischen Prozesse. Insbesondere die möglichen Effekte von Ionen-Partikel-Wechselwirkungen und von Ionenchemie auf die Partikelneubildung werden kontrovers diskutiert. In Ergänzung zu bestehenden Forschungsprogrammen hinsichtlich der Rolle von Ionen im initialen Nukleationsschritt wird vorgeschlagen, Ionen-Partikel-Wechselwirkungen während des anschließenden Partikelwachstums zu untersuchen und sich dabei auf direkte Messungen des Ladungszustands, der Wachstumsraten und der chemischen Zusammensetzung von sekundärem organischem Aerosol zu konzentrieren. Hierzu werden der Ladungszustand und die Wachstumsraten von Partikelpopulationen mit einem modifizierten Mobilitätspartikelspektrometer unter wohldefinierten Randbedingungen in Laborexperimenten quantifiziert. In einem nächsten Schritt werden die neuartigen Messmöglichkeiten unseres Aerosol-Massenspektrometers CAChUP voll ausgeschöpft, um den Beitrag verschiedener organischer Vorläufergase zur chemischen Zusammensetzung von sekundärem organischen Aerosol bei variierenden Ladungszuständen zu quantifizieren. Schließlich werden die Ergebnisse dieser Experimente durch Messungen zur sekundären organischen Partikelbildung bei wohldefinierten Ionenkonzentrationen an einer Aerosolkammer überprüft. Die vorgeschlagene Forschungsagenda ist somit darauf abgestimmt, mögliche ladungs-katalysierte chemische Mechanismen bei der sekundären Aerosolbildung besser einzuordnen.
Various species of pest insects cause substantial damage to agriculture every year, or transmit deadly diseases to animals and humans. A successful strategy to control pest insect populations is based on the Sterile Insect Technique (SIT), which uses the release of mass-reared, radiation sterilized male insects to cause infertile matings and thus reduce the pest population level. However, irradiation is not applicable to every insect species. Thus, new strategies based on genetic modifications of pest insects have been developed or are currently under investigation.The goal of the proposed research is to improve the development and ecological safety of genetically engineered (GE) insects created for enhanced biological control programs, including the SIT and new strategies based on conditional lethality. A major concern for GE insect release programs is transgene stability, and maintenance of their consistent expression. Transgene loss or intra-genomic movement could result in loss of strain attributes, and may ultimately lead to interspecies movement resulting in ecological risks. To address potential transgene instability, a new transposon vector that allows post-integration immobilization will be tested in the Mediterranean, Mexican and Oriental fruit fly tephritid pest species. In addition, the system will be established in the mosquito species Aedes and Anopheles - carriers of dengue and malaria.Random genomic insertion is also problematic for GE strain development due to genomic position effects that suppress transgene expression, and insertional mutations that negatively affect host fitness and viability. Diminished transgene expression could result in the unintended survival of conditional lethal individuals, or the inability to identify them. To target transgene vectors to defined genomic insertion sites having minimal negative effects on gene expression and host fitness, a recombinase-mediated cassette exchange (RMCE) strategy will be developed that. RMCE will also allow for stabilization of the target site, will be tested in tephritid and mosquito species, and will aid to the development of stabilized target-site strains for conditional lethal biocontrol. This will include a molecular and organismal evaluation of an RNAi-based lethality approach. Lethality based on an RNAi mechanism in the proposed insects would increase the species specificity and having multiple targets for lethality versus one target in existing systems. By seeking to improve transgene expressivity and stabilization of transposon-based vector systems, this proposal specifically addresses issues related to new GE insects by reducing their unintended spread after field release, and by limiting the possibilities for transgene introgression.
Das Projekt ÖkoKauf der Stadt Wien hat es sich zum Ziel gesetzt, durch die Erstellung von ökologischen Kriterien, Pilotprojekte und durch Bewusstseinsarbeit das Beschaffungswesen im Magistrat Wien weiter zu ökologisieren. In diesem Rahmen widmete sich der Arbeitskreis 'Desinfektionsmittel unter der Leitung der Wiener Umweltanwaltschaft (WUA) der Aufgabe, für Hygienefachleute ein Instrument zur Beurteilung der Auswirkungen von Desinfektionsmitteln auf Gesundheit und Umwelt zu erstellen. Das Österreichische Ökologie-Institut führte eine Daten- und Literaturrecherche durch, das Umweltbundesamt nahm ergän-zende ökotoxikologische Tests an Wirkstoffen und -produkten vor und 'die umweltberatung ermittelte stationsbezogene Desinfektionsmittelverbräuche in Wiener Krankenanstalten. Die Recherche- und Testergebnisse zu Desinfektionsmittelwirkstoffen und -produkten wurden in einer vom IFZ konzipierten und von der Magistratsabteilung 14 realisierten Datenbank zusammengefasst. Um die ökotoxikologischen Produkteigenschaften vergleichbar zu machen, wurde vom IFZ ein Bewertungsraster entwickelt und in die Datenbank integriert. Dabei werden nachteilige Wirkungen auf die Gesundheit anhand von vier Wirkungskategorien erfasst: Akute Giftigkeit; Reizwirkung auf die Haut; Sensibilisierung, allergenes Potenzial sowie Erbgutschädigende, krebserzeugende und fruchtschädigende Eigenschaften. Zusammen mit der Berücksichtigung des Verhaltens in Oberflächengewässern (Abbauverhalten, Bioakkumulationspotenzial, Toxizität für Wasserorganismen) sowie dem Verhalten in Kläranlagen werden insgesamt sechs Bewertungszahlen generiert, die auf einer Skala von 1 (vernachlässigbar) bis 5 (sehr hoch) das gesamte Gefährdungsprofil des Stoffes beschreiben sollen. Das Gefährdungsprofil eines Handelsproduktes errechnet sich aus den Gefährdungsprofilen der darin enthaltenen Wirkstoffe anhand eines Algorithmus: Dabei wird die Annahme getroffen, dass die Produkteigenschaften von der Konzentration der darin enthaltenen Wirkstoffe abhängen. Bei der Bewertung ist außerdem zu gewährleisten, dass ein Wirkstoff mit einem hohen Gefährdungspotenzial angemessen berücksichtigt wird, auch und gerade wenn seine Konzentration im Produkt gering ist. In der Literatur wird dazu eine logarithmische Skalierung vorgeschlagen. Die Bewertung berücksichtigt derzeit die Wirkstoffe sowie Anwendungsverdünnungen. Die Zusammenfassung der Produkte in Verwendungs- bzw. Expositionskategorien ermöglicht letztlich eine vergleichende Bewertung. Da das Bewertungsraster gerade auf eine vergleichende Bewertung von Produkten abzielt, unterliegt er einer ständigen kritischen Diskussion, die auch häufig von den Herstellern geführt wird. Dieser Umstand sowie das Faktum von Produktlebenszyklen erfordern ein ständiges Update der in der Datenbank enthaltenen Informationen und eine Anpassung des Bewertungsmodells an den aktuellen Stand von Forschung sowie Standards der Stoff- und Produktpolitik.
Shallow groundwater of the huge deltaic systems of Asia like the Red River Delta in Vietnam is often enriched in inorganic arsenic (As), threatening the health of millions of residents. The massive abstraction of groundwater in these areas locally causes an irreversible mixing of arsenic-free groundwater resources with arsenic-rich groundwater. Increased concentrations of competitive anions, especially phosphate (PO43-), decrease the immobilization capacity of the sediments. During transport, the mobility of dissolved As in local aquifers is strongly influenced by adsorption to sedimentary and ubiquitously occurring iron(oxyhydr)oxides. Additionally, arsenic-rich groundwater is often enriched in reduced iron (Fe2+) as well, which is capable to react with iron(oxyhydr)oxides, thereby inducing mineral transformations. Such transformations permanently affect the arsenic adsorption and immobilization capacity of the sediments.Within the scope of this research project, the underlying mechanisms related to As transport and the resulting threat to arsenic-free groundwater resources will be characterized in cooperation with the Swiss Federal Institute of Aquatic Science and Technology (Eawag). The research concept aims at assessing the complex interactions within the arsenic-iron-phosphate-system under field conditions at a study site next to the Red River. First, filtration experiments using local groundwater enriched in As and PO43- will be used to determine the As adsorption capacity of different and previously geochemically characterized iron(oxyhydr)oxides. In a second step, sample carrier containing As loaded iron(oxyhydr)oxides will be introduced into surface near aquifer parts of the study site (via existing groundwater monitoring wells). These samples will be exposed to local groundwater characterized by increased As, Fe2+ and PO43- concentrations for the following nine months. Using the in situ exposition of predefined iron(oxyhydr)oxides, it will be possible to distinguish potential mineral transformations and their influences on the As immobilization capacity of the respective iron(oxyhydr)oxides. By combining the results and outcomes of the field experiments, new and important conclusions regarding the mobility of As can be drawn. The data can be used to create a hydrochemical transport model describing reactive As transport within the investigation area. In addition, the results of the in situ exposition experiments will allow to draw conclusions in respective to the long term As immobilization capacity of different iron(oxyhydr)oxides, which is an essential information regarding in situ decontamination techniques.
The project aims to theorize the scalar organization of natural resource governance in the European Union. This research agenda is inspired by critical geographers' work on the politics of scale. The research will examine an analytical framework derived from theories of institutional change and multi-level govern-ance to fill this theoretical gap. Furthermore, it will review conceptualizations of the state in institutional economics, evaluate their adequacy to capture the role of the state in the dynamics identified, and develop them further. The described processes may imply shifts in administrative levels, shifts in relations between different levels and changes in spatial delimitations of competent jurisdictions that result, for example, from decentralization or the introduction of river basin oriented administrative structures. The research investigates the implications of two European Directives: the Water Framework Directive (WFD) and the Marine Strategy Framework Directive (MSFD). They both have potentially great significance for the organization of marine and water governance at the level of Member States and below, and adhere to similar regulatory ideas for achieving good ecological status of waters. A multiple case study on changes in the scalar reorganization of marine and water governance that result from the implementation of the Directives will be carried out. It will rely on qualitative and quantitative data gathering based on semi-structured interviews and review of secondary and tertiary sources looking at Portugal, Spain, and Germany. It specifically addresses the role of social ecological transactions, the structure of decision making processes and the role of changes in contextual factors (such as ideologies, interdependent institutions and technology).
Beach sand deposits are widespread in the area around Sandefjord, at the western coast of the Oslofjord, southern Norway. The age of the deposits continuously increases with elevation, as the area has been subject to steady glacio-isostatic uplift throughout the Holocene. Existing local sea level curves provide age control related to elevation. Thus, the area offers excellent conditions to test hypotheses on soil formation and OSL dating. A chronosequence covering the last 10 000 years will be established. A preliminary study showed that soil formation leads to Podzols within 4300 - 6600 years. Micromorphological analyses suggest that clay illuviation takes place before and below podzolisation. It is hypothesised that clay translocation goes on contemporarily with podzolisation, but at greater soil depth, where the chemical conditions are suitable. This hypothesis will be proved by more detailed micromorphological investigation and chemical analyses. The factors controlling soil forming processes and their rates, will be determined by analyzing elemental composition, primary minerals and clay mineralogy. Preliminary OSL dating tests suggest that the beach sand deposits are OSL dateable despite the high latitude. This hypothesis will be checked by comparing OSL datings to ages derived from the 14C-based sea level curves.
