Schwerpunktprogramm (SPP) 527: Bereich Infrastruktur - International Ocean Discovery Program, Teilprojekt: Spätpliozen Diatomeenproduktivität in der westlichen Bai von Bengalen: Effekt von Variabilität des Indischen Monsuns und Telekonnektionen

Hauptziel dieses Forschungsverfahren ist die Untersuchung der kieselige Paläoproduktivität in der Bai von Bengalen (Site U1445, ca. 18 Grad N, 84 Grad E) und deren Zusammenhang mit klimatischen, biogeochemischen und hydrographischen Ereignissen, die während dem Spätpliozän geschehen sind. Die Untersuchung der langfristige und die abrupte Schwankungen der Artenzusammensetzung der Diatomeengesellschaft, des Opal und TOC Gehalts und des diatomeenbasierten Isotopensignals, die in dem Zeitraum ca. 3,5 bis 2,5 Myr zu beobachten sind, stehen möglicherweise im Zusammenhang mit der Entwicklung der Klimageschichte des Indischen Monsuns und dem Wechsel zwischen kieseligen und kalkigen Primärproduzenten. Dazu liefern die am Site U1445 erhaltenen diatomeenreichen Sedimente hochauflösende Information über klimatische Geschehnisse und deuten auf herausragende tausendjährige und unter-tausendjährige Schwankungen der ozeanographische Bedingungen im westlichen Indischen Ozean hin. Dieses Forschungsverfahren wird Information über die Phasenregelung der ozeanographische Schwankungen in dem äquatorialem Indischen Ozean beitragen, die von zentraler Bedeutung für unsere Auffassung der Mechanismen hintern des globalen Klimaänderungen sind.

Biologische Monitoringdaten der Oberflächenwasserkörper des Landes Brandenburg

Im Auftrag des Landesamtes für Umwelt (LfU) werden die Oberflächengewässer Brandenburgs biologisch untersucht. Diese Untersuchungen erfolgen alle drei bis sechs Jahre an festgelegten Messstellen. Die für Brandenburg wichtigsten untersuchten biologischen Qualitätskomponenten sind: Phytoplankton (im Wasser freischwebende Algen, zum Beispiel Blaualgen) Diatomeen (schwebende oder am Boden siedelnde Kieselalgen) Makrophyten (zum Beispiel Seerosen, Armleuchteralgen und Wassermoose) Makrozoobenthos (substratgebundene wirbellose Tiere, zum Beispiel Muscheln, Köcherfliegenlarven) Fische Die Datenpakete mit den biologischen Monitoringdaten für die brandenburgischen Oberflächenwasserkörper (für Seen auch physikalisch-chemische Parameter) können aus zusammengestellten Listen (pro Oberflächenwasserkörper ) heruntergeladen werden. Schwerpunktprogramm (SPP) 1158: Antarctic Research with Comparable Investigations in Arctic Sea Ice Areas; Bereich Infrastruktur - Antarktisforschung mit vergleichenden Untersuchungen in arktischen Eisgebieten, Controls on and paleoceanographic utility of the valve size frequency distribution of the Southern Ocean diatom, Fragilariopsis kerguelensis

Unraveling the role of the Southern Ocean's biological pump in regulating climate would be enhanced by the development of paleoceanographic proxies specific to carbon or silica cycling. Observations that the average size of valves of the diatom, Fragilariopsis kerguelensis, varies seasonally, with latitude, and over glacial-interglacial cycles in the Southern Ocean suggest that the valve size frequency distribution in sediments could be used to reconstruct aspects of paleoproductivity and silica cycling. We aim to develop this proxy by using culture, field and sediment samples to determine the exact controls on valve size frequency distributions, the most important of which is likely to be the frequency of auxospore formation within the population. The controls on auxospore formation are as yet unknown and both mating experiments in culture as well as in situ iron fertilization experiments are likely to unravel the mechanisms determining auxospore formation under both controlled and field conditions. The valve size frequency distribution can then be used to indicate the intensity of the environmental and/or biological conditions triggering auxospore formation.

Physiologische, biochemische und biophysikalische Untersuchungen zur Funktion und molekularen Organisation der Xanthophylle im Lichtsammelkomplex der Alge Mantoniella squamata

In diesem Vorhaben wird die Wechselwirkung von Photosynthesepigmenten und Proteinen auf molekularer Ebene untersucht, um besser zu verstehen, unter welchen Bedingungen absorbiertes Licht für die Photosynthese genutzt wird und unter welchen Bedingungen die Energie als Wärmeabstrahlung verloren geht. Ein genaueres Verständnis der zugrunde liegenden Funktionsmechanismen ist z.B. notwendig, um in Zukunft pflanzliche Carotinoide verstärkt in der Biotechnologie einsetzen zu können. In dem Vorhaben wird die molekulare Pigmentorganisation und ihre lichtabhängige Dynamik in einer einzelligen Alge untersucht, die sich durch eine besonders reichhaltige Pigmentierung auszeichnet. Es konnte gezeigt werden, dass der dem Lichtschutz dienende Xanthophyllzyklus bei Diatomeen zusätzlich eine zentrale Rolle in der Regulation der Pigmentbiosynthese spielt. In Mantoniella konnte ein unvollständiger Xanthopyllzyklus nachgewiesen werden. Damit eröffnet sich die Möglichkeit, die Evolution des Lichtschutzes in der Photosynthese experimentell zu untersuchen.

This data set contains the total abundance of the wider taxonomic groups bacillariophyceae, and dinophyceae of the phytoplankton community observed in 2005. Wassergütemessnetz 2 des Landes Brandenburg

Das Wassergütemessnetz 2 (WGMN2) stellt im Rahmen der nationalen und internationalen Meldepflichten aktuelle Daten der interessierten Öffentlichkeit zur Verfügung. Bürger, Schulen und Behörden haben ein reges Interesse an den Daten des WGMN. Deshalb werden die Daten in sechs stationären Gewässergütemessstationen im Zehn-Minuten-Takt aktualisiert. So stehen die erhobenen Parameter in Echtzeit zur Verfügung. Hierbei werden physikalische, hydrologische, meteorologische und biologische Messgrößen erfasst, die eine dynamische Sicht auf die Gewässerbeschaffenheit ermöglichen. Die Messstationen sind an ausgewählten Standorten an der Elbe, Havel, Teltowkanal, Oder und Neiße positioniert. Die Gewässergütemessstationen sind Bestandteil langfristig konzipierter Sanierungsmaßnahmen und dienen dem Nachweis der Gewässergüte und ihrer zeitlichen Veränderung im Rahmen von international abgestimmten Mess- und Untersuchungsprogrammen, der aktuellen Gewässerüberwachung (Warndienste), der Beweissicherung und der Gewinnung von wasserwirtschaftlichen Informationen. Das WGMN trägt dazu bei, dass Auswirkungen von Störfällen bei Industriebetrieben oder von Schiffsunglücken zeitnah ermittelt und zügig Maßnahmen ergriffen werden können. Aber auch kleinere Verunreinigungen wie illegal entsorgtes Altöl vom Auto fallen durch die Messungen schnell auf. Mit der Erkennung von akuten Verschmutzungen und dem Erfassen langfristiger Trends dient das WGMN auch dazu, entsprechende Forderungen der Europäischen Wasserrahmenrichtlinie in Brandenburg umzusetzen. Hier können alle Datensätze abgerufen werden. Derzeit werden die Messwerte im Netz als Grafiken dargestellt.

Nitrate uptake kinetics for Thalassiosira gravida and Thalassiosira rotula

15N nitrate uptake assays were conducted using tracer additions of highly enriched (98%) 15N-labeled nitrate for four Arctic-adapted strains of Thalassiosira gravida and three temperate-adapted strains of Thalassiosira rotula. The experiments were carried out to understand the nitrate uptake kinetics of these strains under controlled laboratory conditions. The conduction of the assays comprised in this dataset took place between 2022-03-09 and 2022-07-15 at the Alfred Wegener Institute, Bremerhaven, Germany. Prior to the experiment, the Arctic strains were maintained at 4°C and the temperate strains at 15°C in climate chambers, with a light intensity of 30 µmol photons m-2s-1 and a photoperiod of 16:8h light:dark. The cultures were kept in exponential growth by semi-continuous dilution with K-medium, enriched with silicon and devoid of ammonium. For precultivation, 2L batch cultures of each strain were grown in ESAW medium at a light intensity of 25 µmol photons m-2 s-1 and a light:dark cycle of 16:8h. Arctic strains were cultivated at 9°C, while temperate strains were grown at 16°C, representing their respective lowest optimum temperatures. The initial nitrate concentration in the medium was set to approximately 100 µM L-1 (equivalent to 1/5 K-medium) to ensure sufficient biomass for subsequent 15N isotope analysis. Upon reaching mid-exponential growth phase, a nitrogen depletion procedure was implemented. This involved decanting 90% of the supernatant from the batch culture flasks and resuspending the sedimented cells in nitrate- and ammonia-free modified ESAW medium. This process was repeated twice more over the course of 48 hours, allowing cells to sediment between each dilution step. This dilution series resulted in a calculated final nitrate concentration of 0.1 µM NaNO3 L-1 at maximum, not accounting for ongoing nitrogen uptake by the diatoms, which likely further depleted the dissolved nitrate concentrations. This approach was deemed the best compromise between maintaining a sufficiently dense culture and achieving nitrogen depletion. 24 hours after the final dilution with nitrogen-free medium, the uptake assay was prepared. Aliquots of 40 ml culture from the batch cultures were transferred into 50 ml culture flasks for the nitrate addition treatments. The experimental design included seven nitrate levels (each with four replicates) and a nitrate-depleted control treatment. The seven nitrate concentrations tested were 0.1, 0.4, 0.8, 2, 10, 50, and 100 µmol NaNO3 L-1. For concentrations of 2 µmol NaNO3 L-1 and below, a 1:1 ratio of 14N:15N was used, while a 9:1 ratio was employed for concentrations above 2 µmol NaNO3 L-1. The experimental units were incubated for 40 minutes under the same conditions as the batch cultures. Following incubation, the samples were filtered onto pre-combusted glass microfiber filters (Whatman GF/F, Maidstone, UK). To reduce dissolved 15N contamination on the filter, an additional 100 ml of nitrate-free medium was filtered through each sample. The filters were then dried at 60°C for 48 hours before being folded into tin capsules and stored in the dark until further analysis. Particulate organic carbon (POC), particulate organic nitrogen (PON), and the particulate 15N:14N ratio were measured using an elemental analyzer (Flash 2000, Thermo Scientific, Karlsruhe, Germany) coupled to an isotope ratio mass spectrometer (IRMS, Thermo Scientific, Karlsruhe, Germany), following the methodology described by Morrien et al. (2017).

Cell densities of the Arctic diatom Thalassiosira gravida in response to temperature, photoperiod and microbiome presence

In March 2023, cell densities of the Arctic diatom Thalassiosira gravida (isolated from the Central Arctic Ocean) were determined to calculate its growth rates at different temperatures and photoperiods in the presence and absence of its natural microbiome. Therefore, a full-factorial experimental design was chosen with two levels of temperature (9°C; 13.5°C) and photoperiod (16h; 24h), to which axenic and xenic diatom cultures were acclimated for one week in climate cabinets prior to the start of the actual growth experiment at a light intensity of 50 µmol photons m-2 s-1. With an initial cell density of 1500 cells/ml, axenic and xenic diatoms were grown under the respective experimental conditions until a cell density of approximately 15000 cells/ml was reached. Cell densities were determined microscopically using an inverted light microscope, following the procedure described in detail in Giesler et al. (2023, 10.3389/fmars.2023.1244639).

KOSMOS 2023 Helgoland mesocosm study on ocean alkalinity enhancement: phytoplankton and microzooplankton abundance and biomass data

This dataset contains species-resolved phytoplankton and microzooplankton abundance and biomass data from the KOSMOS 2023 Helgoland mesocosm experiment conducted in the coastal waters off Helgoland Island in the North Sea, Germany. The experiment investigated the effects of Ocean Alkalinity Enhancement (OAE) on plankton communities during the development of a natural spring bloom. Six mesocosms (~6 m³) were subjected to an immediate dilution OAE scenario, establishing an alkalinity enhancement gradient ranging from ΔTA 0 to 1250 µmol kg⁻¹ in increments of 250 µmol kg⁻¹ through calcium-based alkalinity addition. The dataset includes taxonomically resolved phytoplankton and microzooplankton abundance (cells mL⁻¹), carbon biomass (µg C L⁻¹), functional group classifications, and World Register of Marine Species (WoRMS) taxonomic identifiers. Associated carbonate chemistry measurements, including total alkalinity (TA), dissolved inorganic carbon (DIC), pH, fugacity of carbon dioxide (fCO₂), salinity, temperature, and calculated ΔTA values are provided for each sampling event.
